Cloning Kits

O.D.260 Inc has designed two cloning systems to construct adenovirus vectors:

AdenoQuick™

The AdenoQuickâ„¢ cloning system is ideal for constructing:

  • E1-substituted or E3-substituted adenovirus vectors, especially those difficult to obtain using methods based on plasmid co-transfections and in vitro recombination.
    • Insert your expression cassette either in the E1 region or in the E3 region with the same easiness.
    • Keep the E3 region WT or deleted (2.7 kb).
    • Keep the E4 region WT or deleled (1.2 kb).
    • Maximal cargo capacity is 8.9 kb.
  • Bipartite adenovirus vectors containing two expression cassettes, one in the E1 region, and the other in the E3 region, e.g.:
    • Vectors containing your gene of interest in the E1 region, and a reporter gene in the E3 region, or vice-versa
    • Vectors containing inducible expression systems
    • Vectors expressing a regulator and a regulated gene
    • Vectors expressing two genes involved in the same pathway
  • Oncolytic, conditionally-replicative adenovirus vectors (CrAd's)
    • Clone your tumor-specific promoter in front of the E1a TATA box.
    • Keep the WT E3 region, or insert a therapeutic gene therein ("armed oncolytic vectors").

AdenoZap™

  • AdenoZap™ is one of the fastest systems for constructing E1-substituted adenovirus vectors.
  • It only requires the construction of a small shuttle vector in E. coli before rescuing the virus by DNA transfection. Thus no construction and purification of a large plasmid or cosmid containing the entire sequence of your recombinant adenovirus is necessary.
  • A DNA fragment containing the left ITR, packaging signal, and your expression cassette of interest is purified on agarose gel, and ligated to a ~32 kb-long linear DNA molecule that encompasses the remainder of the adenovirus genome. The ligation product is transfected directly into helper cells (e.g. 293) to rescue the virus, which can form visible plaques as early as 4 days after transfection.
