All recombinant viruses are constructed using our state-of-the-art AdenoZap™ or AdenoQuick™ systems.
Also known as first-generation vectors, these adenoviruses contain one expression cassette in place of the E1 or E3 region. All vectors are E1-deleted. The E3 region is either WT or deleted.
Our AdenoQuick cloning system is the fastest method for constructing recombinant adenoviruses that contain two expression cassettes, one in the E1 region, and the other in the E3 region. This setting is useful for researchers who want to express two genes under the control of two different promoters.
O.D260 has extensive experience in constructing oncolytic viruses also known as Conditionally-replicative-adenovirus (CRAds) that replicate specifically in cancer cells. We have in hand several configurations of the E1a and E1b region that might satisfy your needs. In addition we can easily manipulate the E3 region and the fiber gene via our AdenoQuick cloning system.
O.D.260 has an impressive collection of plasmids carrying all regions of the Ad5 genome. We are able to introduce mutations (deletions, insertions, substitutions..) in any region you wish, reconstitute the genome of your mutant adenovirus using our cosmid technology, and rescue the virus.
It is good microbiological practice to isolate a single virus clone before amplifying it at large scale, even if its entire genome has been assembled in E. coli. We will isolate 3 virus clones by plaque assay and verify their identity.
Get a high-titer preparation of your recombinant virus before you start your in vitro and in vivo studies. We can provide you with highly-purified virus preparation (from 10^12 to 10^15 VP) for a very reasonable price.
You will need to know the titer of your virus prep before you infect cells. Let us do the work for you. We will determine physical titers (virus particles) and biological titers (plaque assay, TCID50, immunostaining....).
Virus preps can be contaminated with replication-competent adenoviruses (RCA) that originate via homologous recombination between the genome of your E1-deleted vector and sequences present in a chromosome of 293 cells. Know your level of contamination before you engage in in vivo studies!
