Construction of Custom Viruses
O.D.260 Inc has extensive experience with manipulating the Ad5 genome. We have a large collection of adenoviral plasmids and are able to reconstitute in large plasmids or cosmids the entire sequence of your recombinant virus containing deletions, substitutions and point mutations at any place (fiber region, E2 and E4 regions…).
Applications:
- Vaccine vectors
- Adenoviruses with modified tropism (mutations in fiber gene…)
- Hybrid Vectors
- Adenoviruses with deletions in essential genes (eg E2, E4), able to replicate only in complementing cell lines
- Adenoviruses with increased packaging capacities
Features:
- Ad5 backbone
- WT or deleted E1 region (3.1 kb, corresponding to bp 354-3503 in the Ad5 genome)
- WT or deleted E3 region (2.7 kb, corresponding to bp 28133-30818 in the Ad5 genome)
The service includes:
- Design of a cloning strategy
- Construction of a plasmid (or cosmid) containing the entire sequence of your specific adenovirus
- Verification of the mutation(s) by sequencing
- Purification of transfection-grade plasmid DNA
- Virus rescue and characterization, unless a specific cell line is required, which OD260 does not have access to

The time required depends on the number of cloning steps necessary. Count on a minimum of 3-4 days per cloning step.
TopAn Important Note on Handling Adenovirus:
Adenovirus is an infectious respiratory virus and its use as a genetic vector requires the use of adequate containment equipment and practices. Biosafety Level 2 (BL2) is appropriate for most customers; however, for some, use of BL3 or higher level containment facility may be required. PLEASE NOTE, O.D.260 Inc. will not carry out projects that require BL3 (or above) containment.
