Construction of Oncolytic Adenoviruses (CRAds)
O.D.260 Inc has extensive experience with construction of oncolytic adenoviruses. We have engineered numerous conditionally replicative adenoviruses (CRAds) for clients around the world.
Applications:
- A genetically engineered virus that can selectively kill cancerous cells while leaving healthy cells intact.
- These engineered adenoviruses have the potential to treat many cancers that are resistant to current treatments. Novel treatment regimes can be created by combining oncolytic adenovirus treatments in conjunction with more traditional treatments like chemotherapy and radiation.

- Possibility of "arming" the virus, i.e. inserting suicide genes, pro-apoptotic genes, etc...
The service includes:
- Design of a cloning strategy
- Construction of a plasmid (or cosmid) containing the entire sequence of your oncolytic adenovirus.
- If necessary, verification of the mutation(s) by sequencing
- Transfection-grade plasmid DNA purification
- Virus rescue by DNA transfection into helper cells (unless these helper cells are specific and unavailable to OD260 Inc.)
- Virus clone isolation
- Confirmation of virus identity at the DNA level

An Important Note on Handling Adenovirus:
Adenovirus is an infectious respiratory virus and its use as a genetic vector requires the use of adequate containment equipment and practices. Biosafety Level 2 (BL2) is appropriate for most customers; however, for some, use of BL3 or higher level containment facility may be required. PLEASE NOTE, O.D.260 Inc. will not carry out projects that require BL3 (or above) containment.
